L’oncialamycine (UCM) appartient à la sous-famille fusionnée à l’anthraquinone des produits naturels enediyne à 10 membres qui présente une cytotoxicité extraordinaire contre un large spectre de lignées cellulaires cancéreuses humaines. Les conjugués anticorps-médicament (ADC), utilisant des analogues synthétiques de l’UCM comme charge utile, sont en développement préclinique. L’UCM est exclusivement produit par Streptomyces uncialis DCA2648 sur milieu gélose solide à faibles titres (∼0,019 mg / L), limitant son apport par fermentation microbienne et entravant ses études de biosynthèse et d’ingénierie par la manipulation des voies in vivo.
Nous rapportons ici les conditions de culture qui permettent la manipulation génétique de la biosynthèse UCM in vivo et permettent la production d’UCM, avec des titres améliorés, par fermentation submergée des souches de S. uncialis modifiées. Ces résultats mettent en évidence une plate-forme biotechnologique pour la biosynthèse, l’ingénierie et la production d’UCM qui devrait faciliter à la fois ses études fondamentales et ses applications translationnelles.
Plus précisément, le titre d’UCM a été amélioré de près de 58 fois à ∼1,1 mg / L grâce à la combinaison de la suppression de grappes de gènes biosynthétiques codant pour des métabolites non liés du type sauvage de S. uncialis, la mutagenèse chimique et la manipulation de régulateurs spécifiques de la voie pour les souches de S. uncialis, et enfin l’optimisation moyenne de ces dernières pour la production d’UCM. La manipulation génétique de la biosynthèse de l’UCM a été démontrée en inactivant des gènes sélectionnés dans les souches modifiées de S. uncialis, dont l’une a donné une souche mutante accumulant la tiancimycine B, un intermédiaire biosynthétique commun connu pour la sous-famille de produits naturels d’énediyne fusionnés à l’anthraquinone.
Applications de la bioluminescence en biotechnologie et au-delà
La bioluminescence est le phénomène naturel fascinant par lequel les créatures vivantes produisent de la lumière. La bioluminescence se produit lorsque l’oxydation d’une luciférine à petite molécule est catalysée par une enzyme luciférase pour former une espèce à l’état excité qui émet de la lumière. Il existe plus de 30 systèmes bioluminescents connus, mais les paires luciférine-luciférase de seulement 11 systèmes ont été caractérisées à ce jour, tandis que d’autres nouveaux systèmes sont actuellement à l’étude. Les différentes paires luciférine-luciférase ont des longueurs d’onde d’émission de lumière différentes et sont donc adaptées à diverses applications.
La dernière décennie environ a vu de grands progrès dans l’ingénierie des protéines, la chimie synthétique et la physique qui ont permis aux luciférines et aux luciférases d’atteindre des applications jusqu’alors inexplorées. La réaction de bioluminescence est désormais couramment utilisée pour les dosages géniques, la détection des interactions protéine-protéine, le criblage à haut débit (HTS) dans la découverte de médicaments, le contrôle de l’hygiène, l’analyse de la pollution dans les écosystèmes et l’imagerie in vivo chez les petits mammifères. S’éloignant de la détection et de l’imagerie, les faits saillants les plus récents des applications de la bioluminescence en biomédecine comprennent le photodécage induit par la bioluminescence de petites molécules
la thérapie photodynamique basée sur la bioluminescence (PDT) et l’utilisation de la bioluminescence pour contrôler les neurones. L’analyse de la génomique fonctionnelle a révélé que les gènes impliqués dans la production de lipides, les canaux ioniques et les antiporteurs sont développés et exprimés. L’exposition au stress thermique modifie le métabolisme des acides gras et augmente la synthèse des acides aminés. L’analyse de co-expression montre que de nombreux gènes de biosynthèse d’acides gras sont surexprimés avec des facteurs de transcription spécifiques sous stress froid. Ces gènes et d’autres impliqués dans la réponse métabolique et régulatrice au stress thermique peuvent être explorés plus avant pour l’amélioration de la souche.
